Job Responsibilities

This role will focus on building and later expanding the in-house liquid chromatography mass spectrometry (LCMS) capabilities for the GTF. Primary responsibilities include, but are not limited to, the following:

• Initiate and execute the development and optimization of LCMS proteomic methods to support AAV and related virus structure-function understanding, including
• Intact mass for capsid protein identification and relative ratios
• Peptide mapping for capsid protein sequence verification and post-translational modifications
• Interrogate regions of the capsid protein for biological function

• Develop and execute a mass spec-based strategy for building and maintaining a protein process residuals data library, including methods to identify and quantitate non-viral proteins from a variety of cell lines used in AAV manufacture
• Function as the LCMS lead on program teams and interact with external stakeholders on Tech Transfer activities, assist in troubleshooting method issues and support validation/qualification campaigns
• Maintain and enhance current knowledge of emerging LCMS methods for virus particles, capsid proteins. nucleic acids and other related molecular entities
• Support improvements for existing LCMS and other biophysical methods
• Represent the company at external events as subject matter expert through presentations and active participation in scientific conferences
• Author and review SOPs, reports and other technical documents and regulatory submissions
• Analyze data with available bioinformatic tools, scientifically-sound interpretation and present the data to internal and external stakeholders.

Qualifications

• PhD / Masters in analytical chemistry, biochemistry or related field, with relevant LCMS proteomics experience in industry and/or academia
• Demonstrated ability to efficiently design, develop and execute LCMS assays from concept to a qualified/pre-validated state, with rigorous attention paid to data processing and analysis
• Extensive hands-on experience in LCMS proteomics techniques including experimental design, data analysis, organization and presentation of complex scientific data
• Demonstrated ability to multi-task and be highly productive in a fluid and fast-paced environment
• A basic understanding of the ICH and USP guidelines for assay qualification and validation
• The candidate should be a self-starter and team player who can balance the needs of an entrepreneurial environment with the requirements of a GMP environment
• Ability to communicate and operate effectively within cross-functional teams
• Proven ability to manage and develop a small team of direct reports

Preferred Qualifications

• Familiarity and/or practical experience with data-independent acquisition (DIA) LCMS methods (SONAR, SWATH, etc.)
• Experience setting up, installing, maintaining and troubleshooting LCMS systems (MS/MS, QTOF, MALDI, Orbitrap, etc.)
• Experience with analytical mass spectrometry (UNIFI/MassLynx, Xcaliber, BioPh) and liquid chromatography (Empower, Chromeleon, OpenLabs, etc.) instrument operation and data processing software environments
• Experience in developing methods and performing the analysis of AAV capsid proteins and/or similar viral vector proteins
• Knowledge and interest in laboratory and equipment automation for high-throughput sample preparation methods
